Cabool is located in south central Missouri at the intersection of U.S. Highways 60 and 63 in the heart of the Ozark Heritage Region. Cabool is only a few miles north of the rugged wilderness of the Mark Twain National Forest and the North Fork River County.

Cabool is located 60 miles south of Rolla, 175 miles southwest of St. Louis, 70 miles east of Springfield, and 235 miles southwest of Kansas City.
